Optimum transfer of a plasmid harbouring the gene for chloramphenicol acetyltransferase (CAT) into protoplasts from cultivated tomato leaves was achieved by electroporation at 666 or 7000 V/cm (single discharge) using a 47 5F capacitor in the presence of 10-100 5g DNA. The CAT gene was strongly expressed when under the control of the CaMV 35S RNA promoter. CAT activity was detected in cells 10 days after electroporation. |
